A flow-through membrane based enzyme immunoassay for the rapid detection of OA in wheat is described. An Immunodyne ABC nembrane was coated with rabbit anti-mouse immunoglobulins and free protein binding sites were blocked. After these antibodycoated membranes were placed on an absorbent layer and a sequential competitive ELISA was performed. The membrane was treated successively with wash solution, monoclonal anti-OA immunoglobulins, wash solution, OA standard solution or sample extract solution, a dilution of OA-horseradish peroxidase conjugate, and wash solution. Finally, H2O2-3,3’,5,5’- tetramethylbenzidine was added for colour reaction. An OA concentration of 0.4 µg/kg resulted in complete colour suppression. With coated membranes, the immunoassay could be performed in less than 15 min.
