Development of a polyclonal antibody-based sensitive enzyme-linked immunosorbent assay for fumonisin B4.

Polyclonal antibodies against FB4, which have good cross-reactivity with four major fuminisins, were produced by jimmunising a rabbit witg FB4-keyhole limpet haemocyanin conjugate and a sensitive competitive direct ELISA for fumonisins was developed. Because of hef limited supplyp of FB4, both FB1-horseradish peroxidase conjugate (HRP) and FB3-HRP were tested as the toxin-enzyme markers in the ELISA. In the FB1-HRP-based ELISA, the coneentrations of FB1, FB2, FB3 and FB4 causing 50% inhibition of binding of enzyme marker (IC50) were 9.0, 2.1, 9.0,and 6.5 ug/L and in the FB3-HRP-based ELISA, the IC50 values were 7.1, 1.9, 7.6 and 5.3 ug/L, respectively. Using the FB3-HRP-based ELISA, the detection limit for FB1 with spiked corn was between 1000 and 500 ug/L. Fusarium moniliforne cultures were analysed with the developed competitive direct ELISA and a well-established FB1 antibody based ELISA, which is not sensitive to FB4. Differences in the fumonisin levels found by the two assays were usde as an indication of the presence of FB4 in the culture material and, therefore, as a method to identify FB4 producing strains.
