A monoclonal antibody based competitive direct ELISA for the rapid analysis of hydrolysed FB1 in corn is described. The concentration required to inhibit enzyme conjugate binding by 50% (IC50) was 36mg/L. The limit of detection of hydrolysed FB1 was 2mg/L, when hydrolysed FB1 was added in boving serum albumin-phosphate buffered saline. The antibody also cross reacted with the hydrolysis products of FB2, B3 and B4, having IC50 values of 331, 174 and 1700 mg/L, respectively. The antibody did not react with intact fumonisins, sphingosine, sphinganine or tricarballylic acid. Samples of corn spiked with hydrolysed FB1 over the range of 5-1000mg/kg indicated the ELISA has a limit of detection of 5mg/kg and an IC50 of 4mg/kg in this matrix.
