pH of cell sap and vacuolar pH during senescence of the rose petal.

The 5 external petals of cut flowers (Rosa hybrida, cultivars Sweet Promise and Royalty) were discarded and measurements were made on the following 5 petals. The pH of cell sap extracted by a freeze-thaw technique was measured. Vacuolar and cytoplasmic pH were measured using radioactive probes. The maximum absorbance wavelength of cell sap was also measured. The pH of cell sap increased with aging in petals, whereas the vacuolar and cytoplasmic pH decreased with aging. Maximum absorbance wavelength of the cell sap increased with increasing pH. It is suggested that the evolution in opposite directions of cell sap pH and vacuolar pH during aging is due to contamination of the cell sap by the cytoplasm during senescence.
