Mode of action of CO2 in delaying senescence of chervil leaves.

Mature chervil (Anthriscus cerefolium) leaves were held in a flow-through system of air or 10% CO2 for 7 days in the dark at 22 deg C. Ethylene inhibitor 2,5-norbornadiene (NBD) was applied in some of the storage jars to yield a final concentration of 2000 mu l/litre. Samples of leaves were taken at 0, 4 and 7 days and analysed for chlorophyll, aldehydes, malondialdehyde and total polyamines. The pH of the cell sap was determined using fresh leaf samples detached at the same time intervals. Chlorophyll degradation and accumulation of aldehydes were delayed in CO2- or CO2 + NBD-treated leaves compared with NBD-treated leaves. CO2 treatment increased the cell sap pH of leaves by one pH unit during 7 days of senescence; cell sap pH was not affected by NBD treatment. This increased pH was due mainly to retention of the original levels of polyamines in CO2- or CO2 + NBD-treated leaves in the course of senescence. During this period, the level of polyamines decreased by 50% in control or NBD-treated leaves. The results indicate that CO2 retards senescence of detached chervil leaves by inhibiting various metabolic senescence-associated processes such as chlorophyll loss and lipid oxidation. In addition, CO2 may retard senescence through its effect on biosynthesis or metabolism of polyamines, thereby maintaining the original content of these senescence retardants.
