Two serological methods to characterize directly Rhizoctonia solani Kuhn AG3 from sclerotia on potato tubers.

A mycelial extract of R. solani AG3 was purified by ion-exchange chromatography. Part of the extract was heat-treated at 60 deg C for 10 min (Ch) and the remainder left untreated (N) prior to chromatography. Antibodies were prepared to a peak fraction present in both extracts and were titrated by immunodiffusion and indirect ELISA. Specificity was examined by immunodiffusion and ELISA of heat-treated extracts of reference strains. Similar results were obtained for the 2 antisera which were both specific to AG3. Characterization tests, using the 2 methods, were carried out directly on sclerotia of tubers of different cultivars and origins harvested in 1991. The purified antigens appeared to be of a complex nature (peptides-polysaccharides) and were very immunogenic. Antibodies raised against these antigens were sufficiently specific as to allow the identification of R. solani AG3 sclerotia on tubers.
