Extracts from ripening tomato fruits contain 3 isoforms of endopolygalacturonase (PG). The 2 lower MW isoforms (PG2 A and B) are heat labile, but the higher MW, thermo-tolerant isoform, PG1, can be formed in vitro from PG2 and a second glycopeptide, a so-called converter molecule. The existence of PG1 as an entity in vivo has been challenged. Alternative hypotheses on the formation and role of PG1 are described and tested and evidence is presented which supports the hypothesis that PG1 exists as such in vivo and may be formed by the interaction of the PG2 polypeptide with an immobile converter peptide.
