ABSTRACT :
Polygalacturonase (PG) produced by C. musae in liquid culture on banana pectic substances as the sole carbon source resolved into 5 distinct isoenzymes, with isoelectric points ranging between pI 4.8 and 6.5. All 5 isoenzymes were detected in banana tissues rotted by C. musae. Isoenzymes PG I (90 kD) and PG II (36 kD) were separated from PG III, IV and V (60-70 kD) by gel filtration on a Superose 12 column. PG III, IV and V were resolved on a hydrophobic column by changing gradients of 0.1 M sodium acetate buffer (pH 5.0). PG II, III, IV and V had endo-activity and PG I had exo-activity. The role of the isoenzymes in postharvest decay of bananas by C. musae is discussed.
