Anticalcifying properties of Dolichos biflorus (horse gram) seeds.

ABSTRACT :
D. biflorus [Macrotyloma uniflorum] is used to treat kidney stones in India and is a constituent of the Ayurvedic medicine, Cystone. The in vitro effects of seed extracts on crystallization of calcium phosphate, a major constituent of kidney stones, were studied. There was a marked decrease in the anticalcifying activity with seed maturation and postharvest storage of seeds for 6 months. The results of the studies suggested that the inhibitors of crystallization present in the seed extract were water soluble, heat stable, polar, non-tannin and non-protein in nature. There appeared to be 2 or more different inhibitors of calcium phosphate precipitation since both the dialysate and the dialysed fractions contained inhibitory activity, although it was more marked for the dialysed fractions. The anticalcifying activity of the aqueous extract was lost completely when it was treated with activated charcoal, and the activity was not recovered or eluted by any solvent tested.
