Identification of chitinase and beta -1,3-glucanase cDNAs from citrus fruit.

Chitinases and beta -1,3-glucanases are well-characterized pathogenesis-related (PR) proteins, and are thought to play a role in the plant's defence responses against fungal pathogens. In order to gain more insight into the molecular mechanisms involved in citrus fruit responses to various stresses, elicitors, and pathogen attacks, this work was aimed at isolating the genes encoding for chitinase and beta -1,3-glucanase from citrus fruit. To achieve these goals, we screened a Valencia flavedo cDNA expression library with Valencia callus chitinase and beta -1,3-endoglucanase antibodies and isolated their corresponding cDNAs. The Valencia flavedo chitinase cDNA, designated chi1, encodes a predicted polypeptide of 231 amino acids and is similar to other grape, maize, rice, and European elder basic chitinases. The Valencia flavedo beta -1,3-glucanase cDNA encodes a predicted polypeptide of 336 amino acids and appears to be identical to gns1, a previously isolated cDNA (GenBank accession no. AJ000081

). The GNS1 polypeptide is also similar to other tomato, potato, soyabean, and tobacco beta -1,3-endoglucanases.
